Wheel nuts are usually torqued up quite tight, and can be difficult to shift especially if you can't put much weight behind them.
That said, unless they've been over tightened or are siezed most people can shift them with the bar that's in the toolkit. If you can't you need to get someone heavier / stronger to have a go, or consider buying an extending wheel brace. These extend to twice the length of the standard brace, so half the weight / strength is required.
Either way you've two options - if you put the brace so that it's horizontal and aligned so that pushing down will undo it, then you can put your weight on the end of the brace.
But if you're not heavy enough to shift it, then put it the opposite way so that an upward lift is required. Then with knees bent, back straight, lift the bar; this time it's your strength working against the car's weight which should move it.

Might seem silly to point this out, but believe me.. some people really do get it wrong... to loosen the nuts you need to turn them anti-clockwise.

this thing you mention "locking wheel nut" it specific to your car, you need this to get the wheel off.
It should be located somewhere in your car, probably the boot.
If you can't find it you will have to go to toyota and then ask them for a new one.
They will need either your chassis number or just your reg.
